UW faculty and staff also enjoy outstanding benefits, professional growth opportunities and unique resources in an environment noted for diversity, intellectual excitement, artistic pursuits, and natural beauty. **The Department of Neurology has an outstanding opportunity for a Research Scientist/Engineer 2 to join their team.** **POSITION PURPOSE** Molecular genetics and gene therapy and find a treatment for muscular dystrophy. **DUTIES AND RESPONSIBILITIES** Mouse Husbandry – 25% + Maintaining a mouse colony (including genotyping, record keeping, interfacing with vivarium staff), planning and executing mouse crosses to generate subject mice for experiments. + Treatment of mice with experimental reagents (e.g. via intraperitoneal, intravenous or subcutaneous injection). + Sacrifice mice and isolation/preparation of tissue samples.Analysis of mice – 25% + Conduct experimental studies on treated and control mice. + Physiology of muscles (measuring mechanical properties such as force development). + DNA and protein quantitation using antibodies, PCR and immunostaining.Molecular Biology – 40% + Maintaining immortalized cells in culture (e.g. HEK293, HeLa, C2, MM14, etc). + Isolation of biomolecules (DNA, RNA and Protein) from tissue and cell samples. + Quantification of target gene expression with quantitative real time PCR (QRT-PCR). + Quantification of target protein expression with western blots. + Staining of tissue slides for immunohistochemistry. + Analysis of immunohistochemical images using existing protocols in the ImageJ software package. + Conducting 96-well micro-plate based assays (e.g. QPCR, Bradford, ELISA, etc.). + Propagation of plasmids, generation of stocks via miniprep or related techniques. + Generation of sequencing libraries (snRNAseq, CUT&RUN, ATACseq, etc). + Cloning and sequencing of plasmids for vector generation (PCR, restriction digests, DNA sequencing, gel electrophoresis).Supervision and training – 10% + Supervise undergraduate student helpers. + Maintain lab training records. **MINIMUM REQUIREMENTS** + Bachelor’s degree in engineering or scientific discipline or a related field and two years of relevant experience. **Equivalent education/experience will substitute for all minimum qualifications except when there are legal requirements, such as a license/certification/registration.** **DESIRED QUALIFICATIONS** + Molecular biology skills (DNA/RNA, cloning, PCR, DNA sequencing). + Experience with antibodies and tissue histology techniques. + Able to read and explain scientific publications. + Experience with small lab animals (mice). + Experience with viral vectors, especially AAV, use and analysis. **Application Process:** The application process may include completion of a variety of online assessments to obtain additional information that will be used in the evaluation process.
